Introduction to Oil-based Delivery Systems

Oil emulsions are often used as a vaccine delivery platform, facilitating the slow release of antigens by forming a reservoir at the injection site. The antigens are trapped in the water phase and are passively released as the emulsion degrades in the body. However, oil-based delivery systems simplify vaccine administration by eliminating the need for emulsion formation.

Oil-based delivery systems can suspend vaccine components directly in the oil thinner, forcing immune cells to actively absorb formulation components without passive release. It can be custom-formulated using a mixture of CD8+ T-cell peptide epitopes, T-helper epitopes, and adjuvants of choice. Liposomes carry hydrophilic antigens and adjuvants directly into the oil medium, capturing all vaccine components in a form suitable for efficient uptake and processing/presentation by antigen-presenting cells (APCs).

There is no need to form an emulsion in a water-free oil-based formulation, as the formulation does not contain water-based ingredients. A strong and long-lasting T-cell immune response can be generated with cancer antigens.

The presence of lipids ensures that all ingredients in the formula are suspended in the oil. In combination with other forms of immunotherapy, the induced active immune response can be further enhanced.
