Oil emulsions are often used as a vaccine delivery platform, facilitating the slow release of antigens by forming a reservoir at the injection site. The antigens are trapped in the water phase and are passively released as the emulsion degrades in the body. However, oil-based delivery systems simplify vaccine administration by eliminating the need for emulsion formation.
Oil-based delivery systems can suspend vaccine components directly in the oil thinner, forcing immune cells to actively absorb formulation components without passive release. It can be custom-formulated using a mixture of CD8+ T-cell peptide epitopes, T-helper epitopes, and adjuvants of choice. Liposomes carry hydrophilic antigens and adjuvants directly into the oil medium, capturing all vaccine components in a form suitable for efficient uptake and processing/presentation by antigen-presenting cells (APCs).
